Abstract:

Obesity-associated metabolic disorders are influenced by gut microbiota, particularly lactic acid bacteria (LAB), through bioactive metabolites such as conjugated linoleic acid (CLA) and branched-chain amino acids (BCAAs). However, these properties are highly strain-dependent. To evaluate local LAB strains for linoleic acid biotransformation, CLA production, BCAA synthesis, and physiological-biochemical safety characteristics. Four strains — Lacticaseibacillus paracasei CM-4.2 and G/S 5, and Pediococcus acidilactici AK 1.2 and AK 1.5 — were studied. CLA was analyzed by GC-MS; free amino acids by HPLC. Safety properties were assessed per MUK 4.2.2602-101. L. paracasei CM-4.2 showed the highest 9, 11-CLA production (0.095 mg/mL vs. control 0.004). P. acidilactici AK 1.2 had the highest precursor 9, 12-CLA (0.0925 mg/mL) and the highest total BCAA content (66.310 mg/mL), especially leucine (34.197 mg/mL). No catalase, lecithinase, gelatinase, amylase, or hemolysin activity was detected. Titratable acidity was 140–142°T; all strains grew at 10°C, but only P. acidilactici strains and L. paracasei G/S 5 grew at 45°C. Results confirm that CLA and BCAA production are strain-specific and cannot be generalized to species level. L. paracasei CM-4.2 shows strong potential for CLA-mediated metabolic modulation, while P. acidilactici AK 1.2 is notable for BCAA synthesis. The absence of virulence factors supports their safety. L. paracasei CM-4.2, P. acidilactici AK 1.2, and P. acidilactici AK 1.5 are promising candidates for metabolically targeted probiotic and synbiotic formulations. Further genomic and in vivo studies are needed.
