Abstract:

The major pigment produced by this bacterium is pyoverdine, a siderophore that has a strong affinity for ferric iron (Fe³⁺. Hence, there is a need for reduction of iron content in domestic water samples using crude pigment extract of Pseudomonas fluorescens. Pigmented bacteria were isolated from collected samples using the serial dilution technique followed by the spread plate method.\Pigmented bacterial isolates were identified based on their fluorescence and pigment characteristics using a UV transilluminator. Pigmented bacterial isolate was evaluated for siderophore production using a P-3,P-4 and Pseudomonas isolation agar. Antimicrobial activity of crude pigmented bacterial isolate by well diffusion method was conducted at various concentrations followed by iron chelation. The analysis of water samples revealed that bore water contained the highest iron concentration (0.456), followed by tap water (0.339) and RO water (0.319). Treatment with siderophore pigment resulted in a substantial reduction in iron levels across all samples. The maximum removal efficiency was observed in bore water, where iron concentration decreased to 0.019, followed by RO water (0.031) and tap water (0.0751).The present study demonstrates that the crude extract of Pseudomonas fluorescens exhibits significant antifungal activity against Aspergillus niger, Aspergillus flavus, Rhizopus spp., and Penicillium spp., with zone of inhibition increasing in a concentration-dependent manner. The highest activity was observed at 500 µL, particularly against Aspergillus flavus (40 mm), followed by Penicillium spp. and Rhizopus spp. (20 mm each). Moderate to low activity was observed at reduced concentrations (250 µL and 125 µL),
