The escalating global threat of antimicrobial resistance (AMR) necessitates the urgent exploration of botanical alternatives rich in bioactive phytochemicals. This study evaluated the preliminary phytochemical composition and antibacterial efficacy of aqueous root extracts from Hibiscus rosa-sinensis L. (Malvaceae) and Lawsonia inermis L. (Lythraceae) against Escherichia coli. Aqueous root extracts were obtained through Soxhlet extraction methods. Qualitative phytochemical screening confirmed the presence of alkaloids, flavonoids, tannins, saponins, and cardiac glycosides in both plant species, with L. inermis exhibiting noticeably higher relative concentrations of tannins and phenolic compounds. Antimicrobial susceptibility testing against Escherichia coli demonstrated that all extract ratios exhibited inhibitory activity, with the equal combination (3:3) producing the largest zone of inhibition (21 mm), followed by 2:3 (14 mm) and 3:2 (12 mm). These findings highlight the complementary phytochemical profiles of Hibiscus and Henna roots and suggest synergistic antimicrobial effects when combined in equal proportions. Minimum Inhibitory Concentration (MIC) values determined via broth microdilution were 12.5 mg/mL for L. inermis and 25.0 mg/mL for H. rosa-sinensis. These empirical findings validate the traditional ethnomedical utility of these botanicals and highlight their potential as novel phyto-therapeutics against Gram-negative enteric pathogens.
