Abstract:

Lignocellulosic biomass represents an abundant, renewable resource for bio-based production; however, its recalcitrant structural matrix limits efficient bioconversion. This study evaluated the biochemical properties and hydrolytic potential of cellulolytic and pectinolytic bacteria isolated from fresh bovine dung. A total of 12 Gram-positive bacterial strains (8 cellulolytic and 4 pectinolytic) were isolated and characterized through morphological and biochemical profiling, demonstrating widespread catalase, urease, and amylase activities alongside diverse fermentative capabilities. Qualitative screening using cellulose Congo-Red and yeast extract pectin (YEP) agar confirmed active extracellular endoglucanase and pectinase production via clear hydrolytic zones. Quantitative enzyme assays revealed significant variation in catalytic efficiency among the isolates. Among cellulolytic strains, isolate BrR displayed the highest endoglucanase activity on amorphous cellulose (0.0080 ± 0.0006 IU/ml), while isolate BrRs exhibited the highest total cellulase activity on microcrystalline filter paper (0.0046 ± 0.0005 IU/ml). A Two-Way ANOVA confirmed that substrate type (F = 1977.36, p < 0.0001), isolate identity (F = 71.93, p < 0.0001), and their interaction (F = 32.73, p < 0.0001) significantly affected cellulase yields. Among pectinolytic strains, isolate PcL3 demonstrated superior pectinase activity (1.52 ± 0.12 µmol.ml-1.min-1) on commercial citrus pectin, releasing 2100 μg of glucose equivalents (F = 66.42, p < 0.0001). These findings highlight isolates BrRs, BrR, and PcL3 as promising biocatalytic candidates for agricultural waste valorization, biopulping, and biofuel production.
