Dengue fever is currently the most important arthropod borne viral disease due to its potential for extensive outbreaks of life-threatening disease. Dengue is an acute viral infection with potentially fatal complications. Early and accurate diagnosis can reduce the mortality & morbidity of the patient. This study aims to assess the various sero diagnostic methods along with molecular characterization of prevalent of dengue serovar (DENV 1-4) in western Rajasthan, A hospital-based cross-sectional study was conducted at Dr. S. N. Medical College, Jodhpur, from January to December 2021. Clinically suspected dengue cases were tested using rapid card test the Dengue Duo rapid card method for NS1 antigen and IgM/IgG antibodies. Dengue IgM MAC ELISA and NS1 antigen ELISA were done using ELISA Kits. RT PCR test were done for molecular characterization of prevalent serovars (DENV 1-4). Out of 12,317 fever cases, 550 (4.47%) were positive for dengue by rapid card testing. Among these, 271 (49.27%) were positive for Dengue IgM ELISA, while 229 (41.64%) tested positive for Dengue NS1 antigen ELISA. The Dengue Duo rapid card test demonstrated a diagnostic accuracy of 50% for IgM and 56.36% for NS1 antigen when compared to ELISA. RTPCR Positivity was seen in – cases and there was predominance of DENV 2 Serovars. Development and commercialization of point-of-care combination tests capable of detecting markers of infection present during different stages of infection (viral nonstructural protein 1 and immunoglobulin M) has greatly simplified laboratory-based dengue diagnosis. By doing RTPCR test prevalent serovars of dengue can be detected which helps in managing DF, DHF & DSS cases.
